Frequently Asked Questions
Q: What is Slope
Spectroscopy?
A: Slope Spectroscopy TM is
at its heart of UV spectroscopy method based upon the Beer-Lambert Law.
However, if differs from traditional spectroscopy methods which hold the
measurement pathlength constant because it varys the measurement
pathlength to collect Absorbance vs. Pathlength or Section data. The
Slope of the Section plot allow rapid quantitation of concentrations and
because the measurement pathlength can change based upon the sample
concentration and absorbance, highly accuract measurements can be made
without the need or cost of dilution steps. To learn more read this
article or
Wiki.
Q: What
concentration ranges can the SoloVPE measure?
A: The SoloVPE System is capable of measureing
a wide range of concentrations, including Protein Concentrations (e.g.
BSA) from below 0.01 mg/ml to over 300 mg/ml and Antibody Concentrations
between 0.01 mg/ml to 250 mg/ml.
Q: How much sample
volume is required to make a measurement?
A: The amount of sample required for a
measurement is dependent on the Sample Vessel being used and the
concentration (which influences the pathlengths to be measured). Highly
concentrated samples can be measured with less than 10 microliters,
however, very dilute samples may required more than 1 ml. In each case
the sample can be recovered
Q: How long does a
concentration measurement take?
A: The fastest and easiest way to measure
concentration with the SoloVPE System is to use the Quick Slope
application. Because Baseline Correction is frequently unnecessary since
the Slope Spectroscopy technique is used, measurement results can
typically be obtain in about 1 minute.
Q: Can the SoloVPE
be used with any spectrophotometer?
A: The SoloVPE System can only be used with
the Varian Cary 50 or Agilent Cary 60. The unique design and
capabilities of those instruments make them ideal for integrating
variable pathlength technology.
Q: Can the SoloVPE
System be used in a GMP Environment?
A: SoloVPE Systems have been installed and
validated in GMP labortatory environments around the globe. When being
installed in a Quality Assurance lab, customers typically purchase the
option security package upgrade call SecureVPE.
Q: Where are SoloVPE
System currently being used?
A: Many companies around the world have
purchased, installed and validated SolovPE Systems. System are installed
in the Americas, Europe and Asia. Many companies have standardized on
the SoloVPE System across multiple sites for concentrations measurements
because of the excellent accuracy and reproducibility of the results.
SoloVPE System have been installed across every functional area from
Discovery and R&D through pilot production, manufacturing and quality
assurance.
